SOP on Operation and Calibration of UV Cabinet

Operation Procedure of UV Cabinet

  • Clean the UV Cabinet from outside and inside with a clean dry cloth.
  • Switch on the main power supply.
  • Open the cabinet door by rotating the knob in either direction.
  • Keep the TLC Plate in U.V. Cabinet.
  • Close the cabinet door by rotating the knob in either direction.
  • Turn “ON” the main switch.
  • Switch on the required UV Light i.e. Short wave (254 nm), Longwave (366 nm).
  • Visualize the plate through an eyepiece from outside the chamber.
  • Switch off the UV Light i.e. Short wave (254 nm), Longwave (366 nm).
  • Turn “OFF” the main switch.
  • Open the cabinet door and remove the plate from the chamber and simultaneously close the door.

Precaution during the handling of UV Cabinet

  • Do not look directly with naked eyes towards the light, when using Long and Short waves.
  • Never work with unprotected hands under UV light, e.g. for marking or adding texts to TLC plates.
  • Use a soft cloth to wipe off the glass from time to time.
  • Before starting ensure that the UV Cabinet is clean.

Calibration of UV Lamp

Frequency of Calibration: 6 Months.

Procedure for a short wave (254 nm) calibration of UV Cabinet

  • Preparation of Sodium salicylate solution(C7H5NaO3)  (Concentration: 0.4 g/L): Accurately weigh and transfer about 100 mg of Sodium salicylate into a 250-mL volumetric flask. Dissolve it insufficient amount of alcohol and dilute to volume with the alcohol.
  • Spot approximately 5 µL of the above solution of Sodium salicylate on the silica gel 60 F254 TLC plate. The spot shall have a diameter of about 5 mm.
  • Allow the spot to dry in the air and examine the Sodium salicylate spot on the TLC plate under Short wave (254 nm).
  • Acceptance Criteria: The lamp shall be capable of revealing the spot on the silica gel 60 F254 TLC plate under a Short wave (254 nm).

Procedure for a Longwave (366 nm) calibration of UV Cabinet

  • Preparation of Sodium salicylate solution (C7H5NaO3) (Concentration: 2 g/L): Accurately weigh and transfer about 200 mg of Sodium salicylate into a 100 mL volumetric flask. Dissolve it in sufficient amount of alcohol, make up the volume with the alcohol.
  • Spot approximately 5 µL of the above solution of Sodium salicylate on the silica gel 60 F254 TLC plate. The spot shall have a diameter of about 5 mm.
  • Allow the spot to dry in air and examine Sodium salicylate spot on the TLC plate under Longwave (366 nm).
  • Acceptance Criteria: The lamp shall be capable of revealing the spot on the silica gel 60 F254 TLC plate under Longwave (366 nm).
